To obtain a precise measurement of gene expression, it is essential to normalize the results of your real time PCR in relation to the fixed expression of the reference gene in such away that experimental conditions do not affect the result.
Although standardization with reference to the constituent expression of the housekeeping gene is the most common method, there is no universal reference gene that is constant in all experimental situations.

For example, the levels of expression of a reference gene can vary for the following reasons:

• The tissue origin of your sample e.g. heart vs. brain
• The disease origin of your sample e.g. carcinoma vs. healthy tissue
• Your experimental parameters e.g. stimulated cells vs. non-stimulated

With a correct standardization strategy, it can be more precise, reproducible and sensitive in your real time PCR.

“Fast selection of reference housekeeping genes”

geNorm is a system for selecting the best candidate reference gene for certain experimental conditions. Simply quantify the 6 or 12 housekeeping genes in your own samples, and together with the geNorm software included in your Kit you will be able to calculate which housekeeping gene is expressed with the most stability in your experimental model.
